    Atrazine induction of a family 4 cytochrome P450 gene in \u3ci\u3eChironomus tentans\u3c/i\u3e (Diptera: Chironomidae)

    Cytochrome P450-dependent microsomal monooxygenase (P450) activity was measured in control and atrazine-exposed third instar midge larvae, Chironomus tentans. Significantly elevated O-demethylase activity was observed in gut homogenates taken from midges exposed to atrazine concentrations from 1 to 10 ppm for 90 h. No significant induction was observed at atrazine concentrations below 1 ppm. A region of a cytochrome P450 family 4 gene was amplified and sequenced from C. tentans larvae. Alignments of inferred amino acid sequences with other insect CYP4 gene homologues indicate a high degree of similarity. Northern blot analysis employing the CYP4 gene fragment as a probe showed an overexpression in C. tentans exposed to atrazine. The results support the previously identified inducibility of cytochrome P450-dependent activity and provide insight into the potential consequences of atrazine exposure to aquatic organisms

    Long-term survival and success of zirconia screw-retained implant-supported prostheses for up to 12 years: a retrospective multicenter study

    Statement of problem: Despite the broad clinical application of zirconia for fixed implant-supported prostheses, evidence of long-term performance is sparse. Purpose: The purpose of this retrospective study was to evaluate the long-term clinical and radiographic outcomes of zirconia-based partial and complete screw-retained implant-supported zirconia fixed dental prostheses (ISZFDPs). Material and methods: Records of patients treated with dental implants and ISZFDPs between December 2004 and June 2017 were screened. Eligible study participants, according to inclusion criteria, were contacted and invited to undergo clinical and radiographic examinations. Outcomes were evaluated as implant and prosthetic survival rates, prosthetic success rate, complications, marginal bone level (MBL) change, and soft tissue condition. Along with the effects of zirconia prosthesis type and level, the effects of implant type and connection, type of loading, and follow-up on MBL were tested with a generalized linear effects model (GLEM) (α=.05). Results: A total of 118 patients were identified, of whom 20 (16.9%) were not available for clinical examination for various reasons. Ninety-eight participants (mean age 60.7 ±11.7 years) with 337 implants were included, of which 176 (52.2%) had been immediately loaded. A total of 111 ISZFDPs (96 zirconia connection and 15 titanium base) were investigated: 24 complete ISZFDPs with a zirconia connection (12.9 ±0.97 dental units, minimum 12, maximum 14), 72 partial with a zirconia connection (3.11 ±1.12, minimum 2, maximum 7), 15 partial with a titanium base (3.62 ±1.02, minimum 2, maximum 5). Forty ISZFDPs had been in function for â‰„10 years (36%), 38 for 5 to 9 years (34.2%), and 33 for 2 to 4 years (22.8%). The mean follow-up time was 7.2 ±3.4 years. No zirconia fractures were identified. Two implants and 2 ISZFDPs failed, with chipping being the most common complication (13.5%). The implant survival rate was 99.4%, and the prosthetic survival rate was 98.2%. The cumulative prosthetic success rate was 91.9%. MBL change was -0.18 ±0.59 mm. Thirteen implants were treated for peri-implantitis (3.8%), and 9 for mucositis (2.7%), but presented healthy peri-implant soft tissues at the follow-up examination. A significant difference was found between the implant-level and abutment-level prostheses (P=.013), with less marginal bone loss observed in ISZFDPs delivered at the implant level. Conclusions: Zirconia-based screw-retained implant-supported prosthesis can be considered a reliable long-term treatment option for partial and complete edentulism. No zirconia fractures were experienced. Stable bone levels and low peri-implantitis rates were reported regardless of the ISZFDP type and level, implant type and connection, and type of loading

    TDT-HET: A new transmission disequilibrium test that incorporates locus heterogeneity into the analysis of family-based association data

    <p>Abstract</p> <p>Background</p> <p>Locus heterogeneity is one of the most documented phenomena in genetics. To date, relatively little work had been done on the development of methods to address locus heterogeneity in genetic association analysis. Motivated by Zhou and Pan's work, we present a mixture model of linked and unlinked trios and develop a statistical method to estimate the probability that a heterozygous parent transmits the disease allele at a di-allelic locus, and the probability that any trio is in the linked group. The purpose here is the development of a test that extends the classic transmission disequilibrium test (<it>TDT</it>) to one that accounts for locus heterogeneity.</p> <p>Results</p> <p>Our simulations suggest that, for sufficiently large sample size (1000 trios) our method has good power to detect association even the proportion of unlinked trios is high (75%). While the median difference (<it>TDT-HET </it>empirical power - <it>TDT </it>empirical power) is approximately 0 for all MOI, there are parameter settings for which the power difference can be substantial. Our multi-locus simulations suggest that our method has good power to detect association as long as the markers are reasonably well-correlated and the genotype relative risk are larger. Results of both single-locus and multi-locus simulations suggest our method maintains the correct type I error rate.</p> <p>Finally, the <it>TDT-HET </it>statistic shows highly significant p-values for most of the idiopathic scoliosis candidate loci, and for some loci, the estimated proportion of unlinked trios approaches or exceeds 50%, suggesting the presence of locus heterogeneity.</p> <p>Conclusions</p> <p>We have developed an extension of the <it>TDT </it>statistic (<it>TDT-HET</it>) that allows for locus heterogeneity among coded trios. Benefits of our method include: estimates of parameters in the presence of heterogeneity, and reasonable power even when the proportion of linked trios is small. Also, we have extended multi-locus methods to <it>TDT-HET </it>and have demonstrated that the empirical power may be high to detect linkage. Last, given that we obtain PPBs, we conjecture that the <it>TDT-HET </it>may be a useful method for correctly identifying linked trios. We anticipate that researchers will find this property increasingly useful as they apply next-generation sequencing data in family based studies.</p

    Protected areas: providing natural solutions to 21st century challenges

    Protected areas remain a cornerstone of global conservation efforts. The double impacts of climate change and biodiversity loss are major threats to achieving the Millennium Development Goals, especially those relating to environmental sustainability, poverty alleviation and food and water security. The growing awareness of the planet’s vulnerability to human driven changes also provides an opportunity to re-emphasize the multiple values of natural ecosystems and the services that they provide. Protected areas, when integrated into landuse plans as part of larger and connected conservation networks, offer practical, tangible solutions to the problems of both species loss and adaptation to climate change. Natural habitats make a significant contribution to mitigation by storing and sequestering carbon in vegetation and soils, and to adaptation by maintaining essential ecosystem services which help societies to respond to, and cope with climate change and other environmental challenges. Many protected areas could be justified on socioeconomic grounds alone yet their multiple goods and services are largely unrecognized in national accounting. This paper argues that there is a convincing case for greater investment in expanded and better-connected protected area systems, under a range of governance and management regimes that are specifically designed to counter the threats of climate change, increased demand and altered patterns of resource use. The new agenda for protected areas requires greater inclusivity of a broader spectrum of actors and rights holders, with growing attention to landscapes and seascapes protected by indigenous peoples, local communities, private owners and other actors which complement conservation areas managed by state agencies. Greater attention also needs to be focused on ways to integrate and mainstream protected areas into sustainable development, including promotion of “green” infrastructure as a strategic part of responses to climate change

    High-order dispersion mapping of an optical fiber

    We report on measurements of high-order dispersion maps of an optical fiber, showing how the ratio between the third and fourth-order dispersion (ÎČ3/ÎČ4) and the zero-dispersion wavelength (λ0) vary along the length of the fiber. Our method is based on Four-Wave Mixing between short pulses derived from an incoherent pump and a weak laser. We find that the variations in the ratio ÎČ3/ÎČ4 are correlated to those in λ0. We present also numerical calculations to illustrate the limits on the spatial resolution of the method.     Whole Organ Engineering: Approaches, Challenges, and Future Directions

    End-stage organ failure remains a leading cause of morbidity and mortality across the globe. The only curative treatment option currently available for patients diagnosed with end-stage organ failure is organ transplantation. However, due to a critical shortage of organs, only a fraction of these patients are able to receive a viable organ transplantation. Those patients fortunate enough to receive a transplant must then be subjected to a lifelong regimen of immunosuppressant drugs. The concept of whole organ engineering offers a promising alternative to organ transplantation that overcomes these limitations. Organ engineering is a discipline that merges developmental biology, anatomy, physiology, and cellular interactions with enabling technologies such as advanced biomaterials and biofabrication to create bioartificial organs that recapitulate native organs in vivo. There have been numerous developments in bioengineering of whole organs over the past two decades. Key technological advancements include (1) methods of whole organ decellularization and recellularization, (2) three-dimensional bioprinting, (3) advanced stem cell technologies, and (4) the ability to genetically modify tissues and cells. These advancements give hope that organ engineering will become a commercial reality in the next decade. In this review article, we describe the foundational principles of whole organ engineering, discuss key technological advances, and provide an overview of current limitations and future directions
